V\u00a0r\u00e1mci CEM SAV bude realizovan\u00e9 hodnotenie l\u00e1tok a\u00a0ich zmes\u00ed z\u00a0h\u013eadiska potenci\u00e1lnej toxicity a\u00a0 imunostimula\u010dn\u00fdch vlastnost\u00ed. S\u00fa\u010dasne sa uskuto\u010dn\u00ed in silico v\u00fdber potenci\u00e1lnych inhib\u00edtorov prote\u00e1zy met\u00f3dou virtu\u00e1lneho skr\u00edningu. V\u00fdber akt\u00edvnych l\u00e1tok pomocou CADD sa bude robi\u0165 na z\u00e1klade virtu\u00e1lneho skr\u00edningu pomocou program MOE. Najprv ohodnot\u00edme z\u00e1kladn\u00e9 skelety chinol\u00ednov, chinol\u00edndi\u00f3nov, pr\u00edpadne benzopyr\u00e1nov pomocou met\u00f3dy molekulov\u00e9ho kotvenia na vybranom modeli SARS-COV-2-3CL prote\u00e1zy z pdb databanky. Po anal\u00fdze interakci\u00ed identifikujeme najd\u00f4le\u017eitej\u0161ie miesta pre derivatiz\u00e1ciu skeletov a vyberieme vhodn\u00e9 substituenty, ktor\u00e9 bud\u00fa z\u00e1kladom pre vytvorenie kombinatorick\u00fdch datab\u00e1z deriv\u00e1tov. Tieto podrob\u00edme virtu\u00e1lnemu skr\u00edningu pomocou multi-kotviacej techniky (multi-docking) a na z\u00e1klade dosiahnut\u00fdch hodn\u00f4t celkov\u00e9ho sk\u00f3re vyberieme set molek\u00fal pre \u010fal\u0161ie \u0161t\u00fadium. Pri hodnoten\u00ed budeme prihliada\u0165 aj na predikovan\u00e9 hodnoty ADMET parametrov z programu ADMET Predictor. Toxicita l\u00e1tok sa bude sledova\u0165 na bunkovej l\u00ednii Vero-6, ktor\u00e1 je zlat\u00fdm \u0161tandardom na \u0161t\u00fadium cytotoxicity kombinovanej s v\u00edrusovou aktivitou. T\u00e1to bunkov\u00e1 l\u00ednia sa pou\u017e\u00edva pre \u0161t\u00fadium replik\u00e1cie, kinetiky, adapt\u00e1cie a cytopatol\u00f3gie SARS-CoV-2 a be\u017ene sa pou\u017e\u00edva aj v\u00a0laborat\u00f3ri\u00e1ch Virologick\u00e9ho \u00fastavu SAV na \u0161t\u00fadie produkcie a infekcie v\u00edrusmi. V s\u00favislosti s v\u00fdskumom SARS-CoV-2\u00a0 bol ned\u00e1vno pop\u00edsan\u00fd pr\u00edstup, ktor\u00fd vyu\u017e\u00edva in vitro trojdimenzion\u00e1lny rekon\u0161tituovan\u00fd tkanivov\u00fd model \u013eudsk\u00fdch d\u00fdchac\u00edch ciest (3D RHuA). In vitro 3D RHuA modely z\u00a0prim\u00e1rnych \u013eudsk\u00fdch buniek maj\u00fa svojimi vlastnos\u0165ami ove\u013ea bli\u017e\u0161ie k \u013eudsk\u00e9mu tkanivu d\u00fdchac\u00edch ciest. S\u00fa kultivovan\u00e9 na rozhran\u00ed vzduch-kvapalina (ALI) a\u00a0v\u010faka ich kultiv\u00e1cii v\u00a0inzert\u00e1ch umo\u017e\u0148uj\u00fa simul\u00e1ciu inhala\u010dnej ako aj syst\u00e9movej expoz\u00edcie. V\u010faka tomu 3D RhuA umo\u017e\u0148uj\u00fa flexibilitu pri modelovan\u00ed fyziologicky relevantn\u00fdch expoz\u00edci\u00ed.\u00a0Modely 3D RHuA, komer\u010dne dostupn\u00e9 ako EpiAirway alebo MucilAir\u2122 maj\u00fa \u0161pecifick\u00e9 mechanizmy na boj proti patog\u00e9nom, ktor\u00e9 s\u00fa porovnate\u013en\u00e9 s\u00a0podmienkami in vivo, v\u00a0\u013eudsk\u00fdch p\u013e\u00facach (napr. produkcia hlienu, mukocili\u00e1rny kl\u00edrens a sekr\u00e9cia obrann\u00fdch l\u00e1tok). Takisto otv\u00e1raj\u00fa mo\u017enos\u0165 \u0161tudova\u0165 infekciu inhal\u00e1ciou alebo syst\u00e9movou cestou.\u00a0 Okrem inhala\u010dnej expoz\u00edcie 3RHuA umo\u017e\u0148uje sledova\u0165 aj syst\u00e9mov\u00fa expoz\u00edciu bazo-later\u00e1lne (t.j. cez kultiva\u010dn\u00e9 medium) v statick\u00fdch ale aj dynamick\u00fdch podmienkach (t.j. v mikro-cirkul\u00e1cii) v\u00a0kultiva\u010dnej komore alebo \u010dipe samostatne alebo v\u00a0kombin\u00e1cii s\u00a0inou 2D alebo 3D kult\u00farou. \u010ea\u013e\u0161\u00ed navrhovan\u00fd model pre pou\u017eitie v\u00a0projekte je 3D rekon\u0161tituovan\u00fd model tenk\u00e9ho \u010dreva, EpiIntestinal, ktor\u00fd je mimoriadne vhodn\u00fd pre \u0161t\u00fadie resorpcie lie\u010div a\u00a0ich aktivity. Podobne ako 3D RHuA je kultivovan\u00fd na ALI a\u00a0umo\u017e\u0148uje \u0161t\u00fadium l\u00e1tok v\u00a0statick\u00fdch aj dynamick\u00fdch podmienkach a\u00a0na multi-tkanivov\u00fdch \u010dipoch. Navrhovan\u00e9 2D a\u00a03D tkanivov\u00e9 kult\u00fary sa pou\u017eij\u00fa na (i) vyhodnotenie toxikologick\u00fdch profilov naj\u00fa\u010dinnej\u0161\u00edch inhib\u00edtorov vybran\u00fdch z in silico modelovania a prote\u00ednov\u00fdch testov; (ii) v\u00fdber necytotoxick\u00fdch nov\u00fdch inhib\u00edtorov, ktor\u00e9 bud\u00fa vhodne pre \u010fal\u0161ie predklinick\u00e9 testovanie. Imunostimula\u010dn\u00e9 vlastnosti l\u00e1tok sa bud\u00fa sledova\u0165 pomocou merania chemoluminiscencie v\u00a0plnej krvi potkana. V\u00a0neutrofiloch, nach\u00e1dzaj\u00facich sa v\u00a0krvi, sa odmeria hladina luminolom potencovanej luminiscencie bez pr\u00edtomnosti a\u00a0v\u00a0pr\u00edtomnosti sledovan\u00fdch l\u00e1tok chrenovej peroxid\u00e1zy.
